RESUMO

The administration of exogenous hormones emerged as an essential tool for field studies in endocrinology. However, working with wild animals remains challenging, because under field conditions not every available method meets the necessary requirements. Achieving a sustained elevation in hormone levels, while simultaneously minimising handling time and invasiveness of the procedure is a difficult task in field endocrinology. Facing this challenge, we have investigated the suitability of biocompatible polymeric microparticles, a novel method for drug-administration, as a tool to manipulate hormones in small songbirds. We chose the insulin-like growth factor-1 (IGF-1) as target hormone, because it receives great interest from the research community due to its important role in shaping life-history traits. Moreover, its short half-life and hydrophilic properties imply a major challenge in finding a suitable method to achieve a sustained, systemic long-term release. To study the release kinetics, we injected either IGF-1 loaded polylactic-co-glycolic acid (PLGA) microparticles or dispersion medium (control group) in the skin pocket of the interscapular region of captive bearded reedlings (Panurus biarmicus). We collected blood samples for 7 consecutive days plus an additional sampling period after two weeks and complemented these with an in vitro experiment. Our results show that in vitro, PLGA microparticles allowed a stable IGF-1 release for more than 15 days, following a burst release at the beginning of the measurement. In vivo, the initial burst was followed by a drop to still elevated levels in circulating IGF-1 until the effect vanished by 16 days post-treatment. This study is the first to describe the use of PLGA-microparticles as a novel tool for exogenous hormone administration in a small passerine. We suggest that this method is highly suitable to achieve the systemic long-term release of hydrophilic hormones with short half-life and reduces overall handling time, as it requires only one subcutaneous injection.

Despite the widely used application of standardized capture-handling protocols to collect blood and assess the physiological stress response, the actual sampling design (e.g., timing and the number of blood samples) often differs between studies, and the potential implications for the measured physiological endpoints remain understudied. We, therefore experimentally tested the effects of repeated handling and multiple blood sampling on the stress response in wintering free-living great tits (Parus major). We modified a well-established sampling protocol of avian studies by adding either an additional blood sample or a "sham-manipulation" (i.e., handling associated with the blood sampling procedure without venepuncture), to disentangle the effects of handling stress and blood loss. We combined three different stress metrics along the endocrine-immune interface to investigate the acute short-term stress response: total corticosterone concentrations (Cort), the heterophil/lymphocyte ratio (H:L), and the Leucocyte Coping Capacity (LCC). Our study provided three key results: i) no relationship between Cort levels, LCC and H:L, confirming that these three parameters represent different physiological endpoints within the stress response; ii) contrasting dynamics in response to stress by the measured parameters and iii) no difference in physiological stress levels 30 min after capture due to one additional blood sampling or handling event. By optimising the sampling design, our results provide implications for animal welfare and planning experimental procedures on stress physiology in passerine species.

Moulting is a crucial, yet often overlooked life-history stage in many animals, when they renew their integumental structures. This life-history stage is an energetically demanding somatic growth event that has particular importance in birds because feathers play a crucial role in flight, insulation and communication. Somatic growth processes are regulated by the evolutionarily conserved peptide hormone insulin-like growth factor 1 (IGF-1). However, the role of IGF-1 in feather growth remains unknown. In this study, we captured 41 juvenile free-living bearded reedlings (Panurus biarmicus) that had started their first complete moult and brought them into captivity. Then, we manipulated their circulating IGF-1 levels using poly-(lactic-co-glycolid acid) microparticles (microspheres) that provide a sustained release of IGF-1. The treatment increased IGF-1 levels but did not affect the feather growth rate. However, 2 weeks after the treatment, birds in the increased IGF-1 group were moulting more feathers simultaneously than the controls and were at a more advanced stage of moult. Birds with experimentally increased IGF-1 levels had better quality feathers (measured by a lower number of fault bars) than the controls. These results suggest that an increase in IGF-1 does not speed up feather growth, but may alter moult intensity by initiating the renewal of several feathers simultaneously. This may shorten the overall moulting time but may imply costs in terms of IGF-1-induced oxidative stress.

OBJECTIVE: To evaluate the incidence and risk factors of generalized convulsive seizure (GCS)-related fractures and injuries during video-EEG monitoring. METHODS: We analyzed all GCSs in patients undergoing video-EEG-monitoring between 2007 and 2019 at epilepsy centers in Frankfurt and Marburg in relation to injuries, falls and accidents associated with GCSs. Data were gathered using video material, EEG material, and a standardized reporting form. RESULTS: A total of 626 GCSs from 411 patients (mean age: 33.6 years; range 3-74 years; 45.0% female) were analyzed. Severe adverse events (SAEs) such as fractures, joint luxation, corneal erosion, and teeth loosening were observed in 13 patients resulting in a risk of 2.1% per GCS (95% CI 1.2-3.4%) and 3.2% per patient (95% CI 1.8-5.2%). Except for a nasal fracture due to a fall onto the face, no SAEs were caused by falls, and all occurred in patients lying in bed without evidence of external trauma. In seven patients, vertebral body compression fractures were confirmed by imaging. This resulted in a risk of 1.1% per GCS (95% CI 0.5-2.2%) and 1.7% per patient (95% CI 0.8-3.3%). These fractures occurred within the tonic phase of a GCS and were accompanied by a characteristic cracking noise. All affected patients reported back pain spontaneously, and an increase in pain on percussion of the affected spine section. CONCLUSIONS: GCSs are associated with a substantial risk of fractures and shoulder dislocations that are not associated with falls. GCSs accompanied by audible cracking, and resulting in back pain, should prompt clinical and imaging evaluations.

PURPOSE: Most patients who die from sudden unexpected death in epilepsy (SUDEP) are found in the prone position. We evaluated whether changes in body position occur during generalized convulsive seizures (GCSs). METHOD: GCSs in patients undergoing video-EEG-monitoring between 2007 and 2017 at epilepsy centers in Frankfurt and Marburg were analyzed in relation to changes in body position. RESULTS: A total of 494 GCSs were analyzed among 327 patients. At seizure onset, positions included supine (48.2 %), right lateral (19.0 %), left lateral (15.6 %), sitting or standing (14.0 %), and prone (3.2 %). Between seizure onset and the start of generalization, 57.5 % of participants altered body positions. During four seizures, patients adopted a prone position, while, in five seizures, patients moved from a prone position. Patients who experienced GCS onset while in a nonprone position had a 2.1 % risk of entering the prone position by the end of their seizure. In contrast, 56.2 % of those in an initial prone position remained so at the end of the GCS, with an odds ratio for maintaining that position of 60.2 (95 % confidence interval: 29.1-124.3; p < 0.001). The likelihood of ending up in the prone position post-GCS did not vary among patients with different nonprone starting positions (p = 0.147). CONCLUSIONS: Seizures in prone position occur during sleep and the highest risk for postictal prone positioning appears to be being in the prone position at GCS onset. Epilepsy patients should therefore be advised to go to sleep in a supine or lateral position to reduce their SUDEP risk.

Avian malaria (caused by Plasmodium spp.) and avian malaria-like infections (caused by Haemoproteus spp.) are widespread and can seriously affect the health of their bird hosts, especially of immunologically naïve individuals. Therefore, these parasites have long been in the focus of bird-parasite studies. However, the species richness and diversity of these protozoan species have only been revealed since the use of molecular techniques. Diversity and prevalence of these parasites among different bird species and even between populations of a species show a large variation. Here, we investigated prevalence of avian malaria and avian malaria-like parasites in two distant populations of a non-migratory wetland specialist passerine, the bearded reedling (Panurus biarmicus). While previous studies have shown that reed-dwelling bird species often carry various blood parasite lineages and the presence of the vectors transmitting Plasmodium and Haemoproteus species has been confirmed from our study sites, prevalence of these parasites was extremely low in our populations. This may either suggest that bearded reedlings may avoid or quickly clear these infections, or these parasites cause high mortality in this species. The remarkably low prevalence of infection in this species is consistent with earlier studies and makes bearded reedlings a possible model organism for investigating the genetic or behavioural adaptations of parasite resistance.

Many organisms use inducible defenses as protection against predators. In animals, inducible defenses may manifest as changes in behavior, morphology, physiology, or life history, and prey species can adjust their defensive responses based on the dangerousness of predators. Analogously, prey may also change the composition and quantity of defensive chemicals when they coexist with different predators, but such predator-induced plasticity in chemical defenses remains elusive in vertebrates. In this study, we investigated whether tadpoles of the common toad (Bufo bufo) adjust their chemical defenses to predation risk in general and specifically to the presence of different predator species; furthermore, we assessed the adaptive value of the induced defense. We reared tadpoles in the presence or absence of one of four caged predator species in a mesocosm experiment, analyzed the composition and quantity of their bufadienolide toxins, and exposed them to free-ranging predators. We found that toad tadpoles did not respond to predation risk by upregulating their bufadienolide synthesis. Fishes and newts consumed only a small percentage of toad tadpoles, suggesting that bufadienolides provided protection against vertebrate predators, irrespective of the rearing environment. Backswimmers consumed toad tadpoles regardless of treatment. Dragonfly larvae were the most voracious predators and consumed more predator-naïve toad tadpoles than tadpoles raised in the presence of dragonfly cues. These results suggest that tadpoles in our experiment had high enough toxin levels for an effective defense against vertebrate predators even in the absence of predator cues. The lack of predator-induced phenotypic plasticity in bufadienolide synthesis may be due to local adaptation for constantly high chemical defense against fishes in the study population and/or due to the high density of conspecifics.

Stress is a collective term for certain conditions and sequences of physiological events enabling living organisms to overcome unpredictable and uncontrollable situations. The context-dependent nature, multidimensional course and large individual variability make stress responses difficult to measure. In avian species, a plethora of studies on short-term stress responses have been conducted by measuring the corticosteroid response to a standardized stress protocol. Here we aimed to test the viability of the leukocyte coping capacity (LCC), measuring oxygen radical production by leukocytes, to assess short-term stress in birds. We collected blood samples from captive house sparrows (Passer domesticus) in the two seasons of winter and spring, right after capture and 30min thereafter. In order to assess the overall physiological stress response to a standardized stressor, i.e. handling and temporary constraint, we measured LCC and additionally combined it with measures of total circulating glucocorticoids (GCs) and oxidative stress. All three methodologies detected significant changes due to the stressor albeit they were not correlated with each other and revealed different information. There was no significant change in oxidative stress levels between the two time points although the amount of relative oxidative damage as well as the anti-oxidative capacity changed significantly. We observed a significant seasonal difference in GC stress response with no difference between sexes. On the contrary, LCC measures revealed with a high individual consistency, that individuals experienced a similar magnitude of stress in both seasons with a significant difference between sexes. Total GC-levels have to be interpreted with caution regarding the assessment of short-term stress reactions. We therefore suggest to supplementary combine classical approaches for measuring stress with the immunological tool of LCC. Our results reveal LCC as a strong and reliable tool to assess short-term stress in captive house sparrows and as promising for other bird species. Collectively the study highlights the necessity to incorporate a range of physiological systems and their endpoints to measure and to assess stress reactions effectively.

Since most avian species have been considered anosmic or microsmatic, olfaction and associated behavioural patterns have hardly been investigated. Most importantly, empirical data on avian olfaction is not equally distributed among species. Initial investigations focused on species with relatively big olfactory bulbs because they were thought to have better olfactory capabilities. Hence, in this study we tested the ability of house sparrows (Passer domesticus) to use chemical cues as parameters to estimate nest features. House sparrows are a commonly used model species, but their olfactory capabilities have not been studied so far. We offered two different odours to males and females, namely the scent of mouse urine (Mus musculus domesticus), representing a possible competitor and a threat to eggs and hatchlings, and the odour of hay, representing a familiar and innocuous odour. The experiment was performed at the sunset to simulate a first inspection to new possible roosting or nesting sites. Interestingly, males but not females preferred to spend significantly more time in front of the hay odour, than in front of the scent of mouse urine. Our results strengthen the hypothesis that oscines can not only perceive odours but also use olfaction to assess the environment and estimate nest site quality.

Amphibians are the most threatened vertebrates today, experiencing worldwide declines. In recent years considerable effort was invested in exposing the causes of these declines. Climate change has been identified as such a cause; however, the expectable effects of predicted milder, shorter winters on hibernation success of temperate-zone Amphibians have remained controversial, mainly due to a lack of controlled experimental studies. Here we present a laboratory experiment, testing the effects of simulated climate change on hibernating juvenile common toads (Bufo bufo). We simulated hibernation conditions by exposing toadlets to current (1.5 °C) or elevated (4.5 °C) hibernation temperatures in combination with current (91 days) or shortened (61 days) hibernation length. We found that a shorter winter and milder hibernation temperature increased survival of toads during hibernation. Furthermore, the increase in temperature and shortening of the cold period had a synergistic positive effect on body mass change during hibernation. Consequently, while climate change may pose severe challenges for amphibians of the temperate zone during their activity period, the negative effects may be dampened by shorter and milder winters experienced during hibernation.

Female song is an ancestral trait in songbirds, yet extant females generally sing less than males. Here, we examine sex differences in the predation cost of singing behaviour. The superb fairy-wren (Malurus cyaneus) is a Southern Hemisphere songbird; males and females provision the brood and produce solo song year-round. Both sexes had higher song rate during the fertile period and lower song rate during incubation and chick feeding. Females were more likely than males to sing close to or inside the nest. For this reason, female but not male song rate predicted egg and nestling predation. This study identifies a high fitness cost of song when a parent bird attends offspring inside a nest and explains gender differences in singing when there are gender differences in parental care.

Do parents defend their offspring whenever necessary, and do self-sacrificing parents really exist? Studies recognized that parent defence is dynamic, mainly depending on the threat predators pose. In this context, parental risk management should consider the threat to themselves and to their offspring. Consequently, the observed defence should be a composite of both risk components. Surprisingly, no study so far has determined the influence of these two threat components on parental decision rules. In a field experiment, we investigated parental risk taking in relation to the threat posed to themselves and their offspring. To disentangle the two threat components, we examined defence behaviours of parent blue tits Cyanistes caeruleus towards three different predators and during different nestling developmental stages. Nest defence strategies in terms of alarm call intensity and nearest predator approach differed between the three predators. Defence intensity was only partly explained by threat level. Most importantly, parental risk management varied in relation to their own, but not offspring risk. Parent defence investment was independent of nestling risk when parents followed a high-risk strategy. However, parents considered nestling as well as parental risk when following a low-risk strategy. Our findings could have general implications for the economy of risk management and decision-making strategies in living beings, including humans.

INTRODUCTION: The differential allocation hypothesis (DAH) predicts that individuals should adjust their parental investment to their current mate's quality. Although in principle the DAH holds for both sexes, male adjustment of parental investment has only been tested in a few experimental studies, revealing contradictory results. We conducted a field experiment to test whether male blue tits (Cyanistes caeruleus) allocate their parental effort in relation to female ornamentation (ultraviolet colouration of the crown), as predicted by the DAH. RESULTS: We reduced the UV reflectance in a sample of females and compared parental care by their mates with that of males paired to sham-manipulated control females. As predicted by the DAH our results demonstrate that males paired with UV-reduced females invested less in feeding effort but did not defend the chicks less than males paired with control females. CONCLUSIONS: To our knowledge, this is one of the first studies providing support for male differential allocation in response to female ornamentation.
